Phenotype
Conditional, Cre-dependent synaptotagmin1 (Syt1) knockout mouse line with loxP sites flanking exon 6 of Syt1. Cre-dependent elimination of Syt1 leads to loss of fast synaptic transmission.

Conditional phenotype: Loss of the exocytotic Ca2+ sensor synaptotagmin 1. Cre-dependent elimination of Syt1 leads to loss of fast synaptic transmission at most synapses. Global elimination of Syt1 is lethal within 48 hrs of birth.

Strain Development
Easi-CRISPR strategy: The donor generated a 635-base ssDNA donor containing a floxed version of exon 6, flanked by two 75-base left and right homology arms. The two parts of the CRISPR guide RNA (crRNA + tracrRNA) and Cas9 protein were combined to generate a ctRNP complex. The long ssDNA donor derived from a floxed exon cassette was mixed with ctRNP(s) to obtain the final Easi-CRISPR reagent cocktail for zygote injection. This was followed by injection of a floxed ssDNA donor with right and left ctRNPs into zygotes resulting in the replacement of the target exon with the floxed exon. Subsequently, microinjection of the Easi-CRISPR reagent cocktail, genotyping, and sequencing were used to identify founders with correctly modified genomes.

If crossed with a global Cre deleter, pups survive less than 48 hrs after birth. Targeting cells and tissues with a more specific Cre recombinase is recommended.
