Genetic Alterations
Random insertion of a transgene where the mouse Vav1 promoter drives the expression of mouse Hottip. The integration position of the transgene has found to be in the intron between exon 3 and exon 4 of the Cdh23 gene. Mice develop anemia, splenomegaly, increased numbers of immature myeloid and die prematurely. These mice may be useful for the study of acute myeloid leukemia.

Strain Development
The Hottip-Tg transgenic construct contains the complete coding region sequence of the mouse Hottip locus under the control of the vav 1 oncogene promoter/control regions (Vav1; from the HS321/45-vav vector). The linearized construct was microinjected into the pronuclei of fertilized C57BL/6J. The transgene integrated into chromosome 10 (Chr10:60117191), within the intron between exon 3 and exon 4 of Cdh23 (cadherin 23 (otocadherin) gene. Founder line 1 was established and maintained on the C57BL/6J background. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (JAX Stock No. 000664).
