Genetic Alterations
This is an inbred strain (IHDID1) developed from the selectively bred line HDID-1 (MMRRC:65277), which was developed to increase the blood ethanol concentration (BEC) achieved after drinking 20% alcohol during a 4-hour period. There is no single allele responsible. Rather, the HDID model is shaped by the influence of many genes.

See MMRRC:65277 for details about selective breeding from the HS/Npt founder population. To generate the inbred line, HDID mice (MMRRC:65277) at selected generation 26 were sib-mated for 22 generations.

This inbred line is one of two genetically independent replicate lines bred for high blood ethanol concentrations (BEC) after a procedure known as "drinking in the dark." This procedure offered the mice a 2-hour period in which to drink 20% v/v ethanol in tap water 3 hours into their dark cycle on Day 1. On Day 2, the period of access to ethanol was extended to 4 hours, with readings of consumption at 2 and at 4 hours. Immediately after reading the final consumption amount, the mice were bled (20 µl) from the periorbital sinus. This blood sample was assayed for BEC by gas chromatography, and the mice of each sex selected for breeding the next generation had the top 15-20 BECs.

At the time of donation, this was the first selective breeding project that used a blood-drug value for the selection index rather than a behavior or physical characteristic. Furthermore, this unique genetic animal model drank to higher blood ethanol concentrations than the standard inbred strain, C57BL/6J, that had been the highest preferring animal model. Importantly, this drinking-in-the-dark model has been adopted by many laboratories to mimic binge-like drinking in mice, and it has been adapted for other rodents (rats, voles, and hamsters). It is simple, minimally invasive, and does not require alcohol adulteration, thirst motivation, long periods of ramping up the ethanol concentration, administration by gavage, or other manipulations. The ethanol tube is placed on the cage of an individually-housed mouse and the water is removed. When the drinking period is ended, water is restored. The mouse may choose whether or not to drink the ethanol.

Embryo cryopreservation was required, as the background "strain" is not an inbred strain, but rather a heterogeneous stock created from an 8-way inbred strain cross, and maintained by a strict, within-family, rotational mating scheme among 48 families (to reduce inbreeding).

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. 2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
