Genetic Alterations
An EGFP-Rpl10a fusion construct was cloned into the multiple cloning regions of a vector containing the promoter and enhancer sequences of endothelin receptor B fused to splicing donor and acceptor sequences of the rabbit β-globin gene containing an intron and polyA sequences using In-Fusion (Clontech) cloning reagents and the primers 5′-AAAGAATTAGCGGCCGCACCATGGTGAGCAAGGGCGAGGA-3′ and 5′-AGTGAATTAGCGGCCGCATACAGACGCTGGGGCTTGC-3′. Founders were screened by PCR using the primers 5′-GCACGACTTCTTCAAGTCCGCCATGCC-3′ (forward) and 5′-CGGATCTTGAAGTTCACCTTGATGCC-3′ (reverse).

Phenotype
Heterozygous Ednrb-EGFP-L10a mice express EGFP-L10a in their enteric nervous system during fetal life, but not during adulthood. They are useful for visualizing and FACS-isolating EGFP+ ENS precursor cells at young fetal ages (i.e., E12.5). This strain has not been bred to homozygosity.

Strain Development
Ednrb-EGFP-L10a mice were made by cloning an EGFP-L10a fusion construct into the multiple cloning region of a vector containing the promoter and enhancer sequences of endothelin receptor B fused to splicing donor and acceptor sequences of the rabbit β-globin gene containing an intron and polyA sequences using In-Fusion (Clontech) cloning reagents and the primers. This vector was injected into the pronucleus of hybrid CBA; C57BL/6 fertilized oocytes. Founders were screened by PCR. Sixteen founder mice were obtained and screened for adult and embryonic ENS expression. A single founder line (line 16) was bred to C57BL/6 mice to produce embryonic mice for these studies. Ednrb-EGFP-L10a transgenic mice were subsequently identified by EGFP fluorescence.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
