Genetic Alterations
Transgenic mice with cardiomyocyte-specific overexpression of human NFE2L2 (a.k.a., NRF2) were generated (Medical University of South Carolina Transgenic Core) in an FVB/NJ background using a transgene cassette containing a murine 5.5-kb alpha myosin heavy chain promoter.

    NRF2 is a transcription factor that regulates the expression of multiple antioxidant and cytoprotective proteins and enzymes, thus playing a key role in protection against oxidative damage. There is a growing interest in understanding the specific role of NRF2 in the development and progression of diseases as well as design novel approaches targeting NRF2 to prevent and/or retard tissue injury. The mice are easy breeders with standard genotyping; they constitute a useful tool in cardiac disease.

    Strain Development

    Transgenic mice with cardiac myocyte-specific overexpression of human NFE2L2 were produced in an FVB/NJ background. Full-length human NFE2L2 cDNA was cloned into a vector containing the alpha-myosin heavy chain (α-MHC) promoter (a gift from Dr. Jeffrey Robbins at Cincinnati Children’s Hospital Medical Center, Cincinnati, Oho) and was used for microinjection at Medical University of South Carolina (MUSC) Transgenic Core. Three founders were identified by PCR and one line with modest overexpression of the randomly inserted construct was established by crossing the founder with wild-type FVB/NJ (RRID:IMSR_JAX:001800) more than 10 times. Mice were then sib-mated for at least 10 generations.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
