Genetic Alterations
This is a novel reporter mouse expressing a fluorescent timer protein (FT-Medium) fused to histone H2B (gene: H2BC9). H2B-FT-Medium is under the control of a doxycycline-inducible promoter (TetO) and knocked into the mouse genome at the HPRT locus on the X chromosome. The mouse also has an rtTA transgene knocked into the Rosa26 locus. This mutant form of rtTA termed M2 has five amino acid substitutions in the tetR moiety of tTA, predicting amino acid exchanges S12G, E19G, A56P, D148E and H179R. Mice are homozygous for the rtTA allele and either homozygous (females) or hemizygous (males) for the iH2B-FT-Medium allele.

Compared to mCherry (RefSeq GenBank AHW57114.1), FT-medium has genetic alterations predicting the following amino acid substitutions: M23E, N28D, T48S, K75R, L88W, M155I, Q193L, L204M, Y219C, R225H.

Strain Development

Correctly targeted blastocysts were implanted into C57BL/6 females. High-degree chimeric male offspring were selected by coat color and backcrossed onto C57BL/6J to N6+. The rtTA allele was introduced by cross-breeding with a strain on C57BL/6J background. Subsequent SNP analysis revealed incomplete backcrossing; the strains genetic background is, hence, a mix of C57BL/6, 129S4, and 129P2.
