Genetic Alterations
These PgR_AF1_QQQ knock-in mice carry genetic variations in the progesterone receptor. The following nucleic acid alterations were introduced, which predicts amino acid substitutions at codons 461, 478 and 489 from KKR (lysine, lysine, arginine) to QQQ (glutamine, glutamine, glutamine). More specifically, K461Q: AAA to CAA, K478Q: AAG to CAG, and R489Q: CGG to CAG.
Phenotype

The progesterone receptor (Pgr) is a nuclear steroid receptor that functions as a ligand activated transcription factor regulating gene expression following progesterone hormone binding. The progesterone receptor gene has two distinct promoters and translation start sites in the first exon which produce two isoforms, A and B. PgR_AF1_QQQ knock-in mice carry K461Q, K478Q and R489Q (lysine, lysine, arginine, respectively to glutamine) substitutions in the activation function 1 domain (AF1) of Pgr. Homozygous knock-in mice are viable and fertile. In cell cultures the QQQ mutations result in hyperactivity of the progesterone receptor isoform B (PRB). This strain may be useful for studying the biology of the mammary gland and uterus.

PgR_AF1_FFF mice carrying the K461F, K478F and R489F mutations are available as MMRRC Stock No. 067160.

Strain Development
CRISPR/Cas9 endonuclease-mediated genome editing was used to create amino acid substitutions at positions 461 (lysine to glutamine, K461Q, AAA to CAA), 478 (lysine to glutamine, K478Q, AAG to CAG), and 489 (arginine to glutamine, R489Q, CGG to CAG) located in the activation function 1 (AF1) domain of the Pgr (progesterone receptor) gene. Single guide RNA and Cas9 endonuclease were introduced into C57BL/6J-derived fertilized eggs by pronuclear injection. Embryos were transferred to pseudopregnant females, and correctly targeted pups were bred to C57BL/6J mice for germline transmission. The mutations were confirmed by sequencing. The colony was maintained on the C57BL/6J background. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
