Phenotype
Homozygous mice were viable. The donor infected wild-type HSV-1 strain McKrae through the eyes, finding that corneal scarring and angiogenesis in the eyes were mild in double-knockouts compared to C57BL/6 and a control Il17ra KO (MRRC:67347).
Note: This line is unique in that it lacks multiple genes from Il17r gene cluster. Il17a is a major member of the Th17 cytokine family and is involved in the pathogenesis of many diseases mediated by inflammation, such as autoimmune disease. Products from genes Il17ra and Il17rc form the major receptor for IL17A. Il17ra is expressed almost ubiquitously, while Il17rc is expressed only in restricted cell subsets; most immune cells do not express Il17rc, raising the question as to whether Il17rc alone or its combination with other IL17R mediate the signal from IL17A. This lines constitutes a valuable tool to dissect the molecular mechanisms of IL17A-mediated signaling events, inflammation, and pathologies.

Strain Development

Double-knockout mice were generated by applying CRISPR genome-editing technology to fertilized eggs of Il17rc-/- mice (B6N.129S5-Il17rctm1Lex/Mmucd, MMRRC:32386). Founder mice were backcrossed twice with C57BL/6J to dilute possible off-target alterations. Homozygous mice were generated by intra-strain mating of heterozygous offspring and maintained by homozygote mating.

This strain of mice is kept in an SPF environment. Since the IL17 pathway is important in immune response, these mice might have unknown deficiencies in immune response and susceptibility to various pathogens.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
