Genetic Alterations
Jup allele: A loxP site was inserted upstream of exon 3. An FRT-flanked neo cassette with a 5' loxP site was inserted downstream of exon 5. Flp-mediated recombination removed the neo cassette and left exons 3 through 5 floxed.

Phenotype
No phenotype without Cre-mediated gene deletion.

The donor reports that, once crossed with Cre-ER, after tamoxifen treatment, all mice are moribund within 10 days. on the other hand, when examined in bone marrow chimeras, deleting both Ctnnb1 and Jup in hematopoietic cells did not affect T cell development, T cell response to viral infection, homeostasis of hematopoietic stem cells, or self-renewal of leukemic stem cells in an acute myeloid leukemia model.

Strain Development
The Jup allele was made using homologous recombination with MM13 ES cells derived from 129S/SvEv. The allele was modified with an unspecified Flp deleted line to remove the neo cassette. The Ctnnb1fl/fl mice were generated using Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) technology. In brief, two LoxP cassettes were inserted into introns 1 and 14, flanking exons 2 to 14 of the Ctnnb1 locus. A mixture containing active guide RNA molecules (gRNA) and Cas-9 protein was injected into the cytoplasm of C57BL/6J embryos. The pups born from the microinjection were screened by PCR, further confirmed by sequencing, and backcrossed to C57BL/6J. The two alleles were combined by crossing the original two strains.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
