Genetic Alterations

The Suz12 conditional allele flanks exons 2 and 3 with loxP sites; it also introduced an Frt-flanked Neo cassette, which was removed through subsequent breeding to a Flp-expressing strain.

    Conditional phenotype: Upon Cre expression, the floxed allele will undergo excision and create a systemic or tissue-specific Suz12 null-allele; an example is described in PMID: 25228648, which used FVB-Tg(Ddx4-cre)1Dcas/J (JAX Strain #006954). The authors determined that Suz12 is required for the viability of male germ cells; see PMID:25228648.

    Strain Development

    To generate Suz12 conditional-ready mice, the donors used a recombineering-based method. The linearized targeting vector DNA was electroporated into E14 cells (derived from 129P2/OlaHsd) subjected to G418/ganciclovir selection. Correctly targeted clones were identified by Southern blot analysis and microinjected into C57BL/6J blastocysts to generate chimeric mice. These chimeras were subsequently crossed with CD-1 females (RRID:IMSR_CRL:022), and heterozygous offspring bearing the targeted alleles were crossed with the Taconic Flp deleter (C57BL/6-Tg(CAG-Flpe)2Arte, RRID:IMSR_TAC:7089) to excise the Frt-bGal-Neo-Frt cassette. This was followed by one cross additional cross to CD-1. Subsequent intrastrain mating for maintenance.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
