Phenotype
Left/right (L-R) axis laterality defects: randomization of abdominal situs, pulmonary right isomerism, vascular heterotaxia, randomized embryo turning, and cardiac looping.

Parallel studies on the partial rescue of zebrafish one-eyed pinhead (oep) mutant embryos showed that by oep mRNA injection resulted in randomization of the direction of heart looping and location of the pancreas, revealing that loss of oep function leads to heterotaxia. Notably, in both Cryptic and oep mutant embryos, L-R asymmetric gene expression did not occur in the lateral plate mesoderm. This might relate to a signaling pathway mediated by Nodal and EGF–CFC proteins that is required for activation of L-R asymmetric gene expression in the lateral plate mesoderm.

Strain Development

A murine Cfc1 cDNA was used to screen a λFIXII library constructed from 129Sv/J genomic DNA (Stratagene), resulting in the isolation of a 21-kb genomic clone containing the entire coding region. A vector containing PGK–neo and PGK–tk cassettes was used to target TC1 ES cells (derived from 129S6/SvEvTac), which were injected into blastocysts. Chimeric males were bred with Taconic Black Swiss females (RRID:IMSR_TAC:sw). After an additional backcross to Black Swiss mice, the line was backcrossed more than 10 generations to C57BL/6NTac.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
