Clathrin conditional-ready knockout mouse model, in which loxP sites were inserted into non-conserved intronic regions enveloping exon 2, predicting a frame-shifting deletion that truncates the protein to 25 amino acids (15 native N-terminal, 10 non-sense frame-shifted residues - MAQILPIRFQEHLQLAKLFRYLTLK*).

Phenotype

These are conditional knockout mice that breed like wild-type mice without Cre recombinase activity. The mice can be crossed with specific Cre mice as per the investigator's region of interest and understand the role of the clathrin gene. Please note that clathrin is essential for development.

Conditional phenotype: When crossed with an amelogenin-driven cre mouse line, the Cltc cK/O mice demonstrate a defect in the tooth enamel prism structure.
Strain Development
For a conditional knockout, the loxP fragments are usually inserted into the introns downstream of the ATG-containing-exon and the removal of the flanked exon(s) will lead to a protein reading frame-shift. After scanning the gene structure and the size of exons, the donor found that exon 2 (ENSMUSE00000997697) can be conditionally removed, predicting a 25-AA product (15 native N-terminal amino acids, 10 frame-shift AAs) that may be subject to non-sense mediated decay. To this end, the donor used the Cre-loxP system, targeting introns 1 and 2, which are big, such that insertion of loxP elements will not interfere with mRNA splicing. Microinjection into C57BL/6N zygotes. To confirm positive F0 founders, PCR and sequencing was performed. Positive F1 mice were validated in 3 step: PCR, Southern analysis, and sequencing.

The line was sib-mated for three generations.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
