Genetic Alterations
Knock-in of NLS-lacZ into the first exon of the Mds1 isoform of the mouse Mecom locus, resulting in a null allele; LacZ is under the transcriptional control of the Mds1 promoter. The construct was assembled in a pBluescriptbackbone containing the following components: the Herpes simplex virus thymidine kinase gene, a lacZ encoding β galactosidase with a nuclear localization signal and a bovine growth hormone (bGH) polyadenylation site; and a neomycin selectable marker with a PGK promoter and pGH polyA site. The lacZ marker was inserted immediately downstream of the first ATG in exon 1 of Mds1, but upstream of the splice donor at the end of the exon; the lacZ insertion is situated such that the splice donor for exon 1 is lost.

Phenotype
The Mds1-lacZ strain is fertile with normal longevity and no discernable phenotype as a heterozygote. Homozygotes present with decreased fecundity due to kyphosis, but the mice breed. Longevity is decreased. There is severe and progressive kyphoscoliosis beginning at 3 weeks, accompanied by vertebral synostosis and osteopenia. The marrow elements are normal but the number of hematopoietic stem cells is markedly diminished (flow cytometry) or absent (bone marrow transplantation of irradiated mice). The Mds1-lacZ allele allows for X-gal staining of gene expression, which, in the embryo, includes hematopoietic stem cells (only in the dorsal aorta, not the fetal liver), mesonephros, limbs, and parts of the brain. This gene serves as a unique and specific marker of HSC emergence in the endothelium-to-hematopoietic transition.

Note: The other isoform of Mecom, Evi1, is expressed at normal levels.
Strain Development
The plasmid was purified and electroporated into TC-1 embryonic stem cells derived from 129S6/SvEvTac. Gancyclovir/G412 and G418-resistant colonies were screened by Southern blot; cells showing the appropriate restriction enzyme polymorphism were injected into C57BL/6-derived mouse blastocysts. Offspring with a high degree of coat chimerism were mated to C57BL/6 mice to test for transmission of the knockout allele.
