Genetic Alterations
CRISPR/Cas-9 induced insertion of two loxP sites - one loxP site 172 base pairs upstream of ORF base position 124 in intron 1, and one loxP site 182 base pairs downstream of ORF base position 371 in intron 2 of the Mmp20 gene on Chromosome 9. In the presence of Cre recombinase, this insertion is predicted to induce the deletion of 636 base pairs: one of the loxP sites and the 602 base pairs between the loxP sites which includes ORF base positions 124-371, and prevents the transcription and translation of the Mmp20 gene.
Phenotype
The phenotype of homozygotes and hemizygotes has not been investigated.
Strain Development

Single-stranded template DNA and guide RNAs were microinjected into C57BL/6J embryos and implanted into foster mothers to result in gene-edited F0 mice. F0 mice with the desired mutation were mated to animals that harbored the Cre deleter allele (Tg(KRT14-cre/ERT2)1Ipc - MGI:2177426) and the mTmG reporter allele (Gt(ROSA)26Sortm4(ACTB-tdTomato,-EGFP)Luo - MGI:3716464) of uncertain genetic background (“K14-CreER; mTmG” in PMID:11034212 and 17868096) to produce the F1 generation. F1 animals that did not harbor the K14-CreER allele were crossed with B6J(FVB)-congenic beta-actin cre mice (Tmem163Tg(ACTB-cre)2Mrt - MGI:2176050) to test if the floxed allele can recombine successfully. Cre-negative F2 progeny were sib-mated thereafter, and the mTmG allele was bred out.
