Genetic Alterations
CRISPR/Cas9 induced deletion of ORF base positions 4-64 and two base pairs downstream of ORF base position 64 in intron 2, and insertion of a Cre recombinase and an SV40 polyadenylation sequence after ORF base position 3 of the Klk4 gene on the 9th chromosome. This alteration is predicted to induce the expression of Cre recombinase instead of the Klk4 protein in cells where and when the Klk4 gene is normally expressed.
Phenotype
Hom: Homozygous KLK4-Cre mice express RFP in maturation-stage ameloblasts and sporadically in the mesenchyme when combined with an RFP reporter allele.
Het/Hemi: Heterozygous KLK4-Cre mice start expressing Cre recombinase in maturation stage ameloblasts.
Strain Development

Single-stranded template DNA and guide RNAs were directly microinjected into C57BL/6J embryos and implanted into foster mothers to result in gene-edited F0 mice. F0 mice with the desired mutation were mated with homozygous B6.Cg-Gt(ROSA)26Sortm14(CAG-tdTomato)Hze/J (RRID:IMSR_JAX:007914) mice to produce F1 mice, which were mated with each other to establish a colony. F2 mice that did not harbor the tdTomato allele were mated with B6.129P2-Gt(ROSA)26Sortm1(CAG-Brainbow2.1)Cle/J (“R26R-Confetti”) and F3 mice were crossed to C57BL/6J mice for at least one generation to cross out the R26R-Confetti allele prior to archival at the MMRRC.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
