Phenotype
Msx2 deficient mice have defects of skull ossification and persistent calvarial foramen. Msx2 mutant mice also display defective tooth, hair follicle and mammary gland development, and seisures, the latter accompanied by abnormal development of the cerebellum.

Strain Development
Fertilized eggs for microinjection were harvested from superovulated 6-week-old female F1(C57B1/6J x CBA/J) mice impregnated by adult male F1(C57/6J x CBA/J) mice. Progeny were analyzed for transgene status by Southern blot hybridization of DNA isolated and hybridized to random primed 32P-labeled luciferase cDNA. Transgene positive mice were bred backcrossed three generations to BALB/c and showed similar transgene expression patterns as reported in the reference below and previously. The line was further backcrossed for at least an additional five generations to BALB/c to ensure that the SEM and TEM studies would be performed on animals homozygous for the transgene.

    * Homozygous knockout mice must be fed a liquid diet; Bio-Serve cat#F1264SP is recommended. This is necessary for mutant husbandry due to tooth phenotype.
    ** MMRRC has noticed lower fertlization rates in MSX2 as compared to MSX1 during IVF procedures, and lower numbers of MSX2 pups born.

      1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

      2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

      3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
