Strain Information

Klk4em4Odk
Name: kallikrein related-peptidase 4 (prostase, enamel matrix, prostate); endonuclease-mediated mutation 4, Ophir D Klein
Synonyms: KLK4-MCHERRY
Type: Allele
Species: Mus musculus (mouse)
Chromosome: 7
Alteration at locus: CRISPR
Klk4
Name: kallikrein related-peptidase 4 (prostase, enamel matrix, prostate)
Synonyms: Prss17, KLK-L1, ESMP1
Type: Gene
Species: Mouse
Chromosome: 7
Alteration at locus: CRISPR
NCBI: 56640
HGNC: HGNC:6365
Homologene: 55856
Genetic Alterations
CRISPR/Cas9 induced deletion of ORF base positions 4-64 and two base pairs downstream of ORF base position 64 in intron 2, and insertion of a gene for mcherry fluorescent protein, and a SV40 polyadenylation sequence after ORF base position 3 of the Klk4 gene on the 9th chromosome. This alteration is predicted to induce the expression of mcherry fluorescent protein instead of the Klk4 protein in cells where and when the Klk4 gene is normally expressed.
Phenotype
Hom:The phenotype of this homozygote has not been investigated.Het/Hemi: Heterozygous KLK4-mcherry mice express mcherry in maturation stage ameloblasts.
Strain Development
Single-stranded template DNA and guide RNAs were directly microinjected into C57B6/J embryos and implanted into foster mothers to result in gene-edited F0 mice. F0 mice with the desired mutation were mated with C57B6/J mice to produce F1 mice, which were mated again with C57B6/J mice to establish a colony. Males were cryopreserved as sperm, available for depositing into the repository.
Suggested Control Mice
C57BL/6J mice
MMRRC Genetic QC Summary
The MMRRC Centers have developed a genetic QC pipeline using MiniMUGA array genotyping to provide additional information on strain backgrounds for MMRRC congenic and inbred strains. For more information on when data may be available, or to request genotyping for a strain of interest, please contact mmrrc@ucdavis.edu. Older strains may not have this information.
Donor
Ophir Klein, M.D., University of California, San Francisco.
