Genetic Alterations
A BAC clone (RP21-11805) containing the mouse Pou5f1 gene was modified via standard BAC recombineering to contain rtTA-advanced (aka rtTA.M2, hereafter simply referred to as rtTA) led by a KOZAK translation initiation sequence, and a bGHpA polyadenylation sequence at the initiating ATG codon of the first coding exon of Pou5f1.

Phenotype

rtTA-advanced (aka rtTA.M2) under the control of the Pou5f1 (Oct4) promoter in inner cell mass of blastocyst, endoderm of epiblast, and E7.5 embryo and extra-embryonic tissue. The allele also causes the development of a "kinky tail" visible as early as E13.5, which is of interest in spine research.

Strain Development
The BAC transgene was linearized and injected into the pronucleus of fertilized F2 mouse embryos from the B6SJLF1/J strain (RRID:IMSR_JAX:100012) using standard microinjection methods. Biopsies were collected from progeny before weaning to identify founders by PCR. Subsequent outcross to CD1. Import to the MMRRC used C57BL/6J.
