Genetic Alterations
A targeting vector was designed to insert a loxP- and FRT-flanked neomycin resistance (neo) cassette upstream of exon 5, and a second loxP site downstream of exon 8. FLPe-mediated recombination removed the FRT-flanked cassette leaving exons 5 through 8 floxed. Global Cre-mediated recombination removed exons 5 through 8. Exons 5 through 8 encode a substantial portion of the catalytic domain of the gene.

Strain Development
Following electroporation into iTL-IC1 embryonic stem cells derived from C57BL/6NTac, correctly targeted ESCs were injected into BALB/c blastocysts to generate chimeras, which were crossed to C57BL/6 females to obtain F1 progeny. These Adamts9fl-Neo/+ mice were crossed with B6.Cg-Tg(ACTFLPe)9205Dym/J (RRID:IMSR_JAX:005703) to excise the neo selection cassette. This was followed by a cross to wild-type C57BL/6 for germline transmission of the Neo-deleted floxed allele and elimination of the ACTB-FLPe transgene. Intercrosses of Adamts9fl/+ mice provided Adamts9fl/fl mice in the expected Mendelian ratio. These mice were viable, fertile and externally normal when followed for up to 1 year of age, suggesting that the inserted loxP sites did not interfere with Adamts9 function. In particular, Adamts9fl/fl mice lacked the highly penetrant, externally evident ocular phenotype reported in Adamts9lacZ/+ mice (PMID:20093484). The mice were mated with B6Ei.129S4-Tg(Prm-cre)58Og/EiJ (RRID:IMSR_JAX:007252) for deletion of Adamts9 in the male germline. Male mice carrying both the Prm1-Cre and Adamts9fl transgenes were crossed with female mice to obtain mice with a germline-deleted allele and lacking the Prm1-Cre transgene. The line is C57BL/6J-congenic (N21) with the Y chromosome fixed to C57BL/6J.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
