The peroxisomal biogenesis factor 10 gene (Pex10) encodes a peroxisomal biogenesis factor 10 protein. Mutations in this gene have been associated with Zellweger syndrome and peroxisome biogenesis disorders type 6A and 6B.

To create the Pex10C286C,G287G,H288D allele, CRISPR/Cas9 endonuclease-mediated genome editing of (Pex10) exon5 was used to introduce a H288D missense mutation that corresponds to the human H310D clinical mutation associated with Zellweger syndrome and peroxisome biogenesis disorders type 6A and 6B. If additional characterization of these Pex10 C286C,G287G,H288D mice is performed, we may modify the strain description accordingly.

Heterozygous mice are viable and fertile. Although homozygous mice are generated in a Mendelian ratio, they are small and inactive relative to heterozygous or wild type littermates.

Strain Development
The Pex10em1 allele was generated using CRISPR/Cas9 endonuclease-mediated genome editing. Guide RNAs were selected to target exon 5 of the peroxisomal biogenesis factor 10 gene (Pex10). Single stranded oligonucleotide donor DNAs 5'-TAGGAGTTCCCTTGAAGACAGAGCCGTGTGCAGAACCCCACTCTGCACCCTATGCTTAGAGGAACGAAGACACTCCACGGCCACACCTTGCGGTGATCTCTTCTGCTGGGAGTGCATCACCGAGTGG-3' were used in combination with guide AGCAGAAGAGATGGCCACA to generate the H288D missense mutation. (CAT->GAT). These sequences and Cas9 nuclease were introduced into single cell C57BL/6J zygotes and transferred to pseudopregnant females. Additionally, the following silent nucleotide changes were included to improve efficiency by preventing guide reassociation and retargeting of the modified allele with CRISPR/Cas9 (C286C [TGT->TGC] and G287G [GGC->GGT]). Progeny were screened by DNA sequencing of Pex10 exon 5 to identify correctly targeted pups, which were then bred to C57BL/6J mice (Stock No. 000664) for germline transmission. The Pex10em1 colony was backcrossed to C57BL/6J for at least two generations.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
