Bard1-D488H knock-in mice express a D488H (aspartic acid to histidine) amino acid substitution in the Bard1 (BRCA1 associated RING domain 1) locus. The D488H mutation is homozygous embryonic lethal. This strain may be useful in studies of tumor suppression and breast cancer.

Phenotype
Bard1 (BRCA1 associated RING domain 1) encodes a protein that interacts with the N-terminal region of BRCA1 and has homology with the 2 most conserved regions of BRCA1, the N-terminal RING motif and the C-terminal BRCT domain. The BARD1/BRCA1 interaction is implicated in tumor suppression. Mutations in BARD1 are associated with breast cancer and hereditary breast ovarian cancer syndrome.

Bard1-D488H knock-in mice express a D488H (aspartic acid to histidine) amino acid substitution. The mutation corresponds to human tumor-associated missense mutations in the ankyrin domain that disrupts BARD1 recognition of H4K20me0 (histone H4 unmethylated at lysine 20). Mice heterozygous for the mutation are viable and fertile. Embryos homozygous for the mutation are not viable. This strain may be useful in studies of tumor suppression and breast cancer.

Strain Development
The targeting vector contains a loxP-flanked neomycin inserted into intron 6 and an aspartic acid to histidine amino acid substitution at position 488 (D488H; GAT to CAT) inserted into exon 6 the Bard1 (BRCA1 associated RING domain 1) gene on chromosome 1. The construct was electroporated into (C57BL/6J x 129S6/SvEvTac)F1-derived KV1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric male was crossed to B6.FVB-Tg(EIIa-cre)C5379Lmgd/J to remove the neo cassette. The strain has been backcrossed to C57BL/6J for two generations. Upon arrival, mice were bred to C57BL/6J for at least 1 generation to establish the colony.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
