Genetic Alterations

This strain harbors a CRISPR-generated conditional mutant of the P2rx3 gene. LoxP sites were inserted on both sides of exon 2. Recombination leads to a frameshift and premature STOP codon in exon 3.

Phenotype
Conditional loss of P2rx3 in mouse auditory neurons leads to altered patterns of neuronal spontaneous activity and neuron differentiation defects. This mouse line may also be useful for studying P2RX3 function in sensory perception and pain modulation.

Conditional phenotype: Mice carrying two copies of the allele are viable. The strain may have conditional phenotypes when crossed to a cre-expressing strain.

Strain Development
P2rx3 floxed mice were generated by CRISPR-mediated insertion of a construct containing loxP sites on either side of exon 2. Injections were done on B6SJLF2 single-cell embryos. Following validation (by PCR), the mice were backcrossed to C57BL/6 for one generation then inbred for an additional generation.
Suggested Control Mice
P2rx3 floxed homozygotes that do not carry Cre recombinase show normal levels of P2RX3. So, P2rx3 fl/fl or fl/+ mice without Cre could be used as controls. But, littermate Cre-positive P2rx3+/+ mice should be used if possible given possible side-effects of Cre lines.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.

4 The distribution fee covers the expense of resuscitating mice from the cryo-archive; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary.
