Genetic Alterations

A transgenic construct containing the protein coding region of the mouse sodium channel, nonvoltage-gated 1 beta gene (Scnn1b) under the control of the rat secretoglobin, family 1A, member 1 (uteroglobin), Scgb1a1, promoter and SV40 polyadenylation site sequence was injected into fertilized B6C3F1 donor eggs. Founder line 6608 was established.

    Phenotype
    The phenotype of this strain (which spontaneously developed during the backcrossing of the Scnn1b-Tg line into a BALB/cJ background; founder line 6608) was noted to have dramatically improved survival rates as compared to the “normal” backcrossed lines. Backcrossing to BALB/cJ produced a high mortality rate as compared to the originally mixed strain. Interestingly, this strain (MMRRC:69779) displays a high survival rate. It was discovered that the improved survival rate was due to a lack of transgene overexpression during the early postnatal period. This resulted in less mucus plugging during the neonatal period accompanied by a lack of emphysema development, which is a feature normally associated with transgene overexpression. Nonetheless, in the adult mice, robust mucus plugging still occurred and this was deemed to be a useful phenotype to maintain and characterize.
    Strain Development

    A transgenic construct containing the protein coding region of the mouse sodium channel, nonvoltage-gated 1 beta gene (Scnn1b) under the control of the rat secretoglobin, family 1A, member 1 (uteroglobin), Scgb1a1, promoter and SV40 polyadenylation site sequence was injected into fertilized B6C3F1 donor eggs. Founder line 6608 was established. Scnn1b transgenic mice were then taken and crossed with BALB/cJ background for approximately 12 generations.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
