Genetic Alterations
Exon 9 of Sh2b1 gene was deleted using CRISPR/Cas9 technology. Sh2b1 gene encodes 4 known isoforms (alpha, beta, gamma, delta) that differ only in their N-termini. Deleting exon 9 results in the deletion of the ubiquitously expressed beta and gamma isoforms. The more restrictively expressed (primary brain) alpha and delta isoforms are still expressed. Tissues that are in WT mice express only the beta and gamma isoforms start expressing the alpha and/or delta isoforms.

Phenotype
The male Sh2b1 beta/gamma KO mice exhibit increased weight, food intake, and adiposity. Insulin and glucose tolerance were normal in both male and female mice. However, fed insulin levels were elevated in the female homozygous mice. The heterozygous mice breed normally and the homozygotes tend to be mildly aggressive.

Strain Development
Fertilized C57BL/6J x SJL F2 oocytes were injected with guide RNAs and oligo template and implanted into C57BL/6J x SJL F2 dams. Mice were backcrossed to C57BL/6J mice for 6 generations. The genotype of the founder mouse was confirmed via PCR and sequencing

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
