Phenotype
This allele has the same phenotype as the original spontaneous Sey allele on the C57BL/6J background. No known observations have been found to have any impact with the addition of the 3xFLAG at the ATG. This is expected since the allele is not expressed as protein until a therapy is attempted.

This strain was specifically made for development and quantification of the efficiency of gene-editing (CRISPR) for aniridia using the Sey allele, because it allows for the differential detection of protein from each allele (antibodies can not distinguish them). Successful use of this strain for this purpose is reported in PMID:32258211. Also listed are other therapeutic approaches this strain will be useful for:

  1. Small-molecule nonsense suppression therapy,
  2. CRISPR/Cas13 strategies that target Pax6 mutations at the RNA level without permanently altering the genome
  3. Mutation-independent strategies (e.g., intronic insertion of Pax6 cDNA sequence immediately downstream of the AUG)
  4. Use of the spliceosome-mediated RNA trans-splicing (SMaRT) technique

Strain Development
This is the same allele as MMRRC 066963-MU, but bred to the 129 background.
Zygotes from 0.5 post-coitus plugs were obtained by the superovulation of C57BL/6J females mated with B6-Pax6Sey/+ males.Optimized CRISPR components were microinjected into one-cell zygotes and implanted into pseudopregnant CD-1 females, producing 24 potential founders. PCR-based genotyping and Sanger sequencing confirmed presence of the transgene in female Founder #7, which was mated to C57BL/6J.The progeny showed stable transmission of the transgene. A male progeny with the transgene was selected as the first-pedigree mouse for further breeding and expansion. The line was backcrossed to C57BL/6J five times. Then, was backcrossed to 129S1 for seven generations.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
