Strain Information

Tmem106btm2d(KOMP)Wtsi
Name: transmembrane protein 106B; targeted mutation 2d, Wellcome Trust Sanger Institute
Type: Allele
Species: Mus musculus (mouse)
Chromosome: 6
Alteration at locus: Knockout
Tmem106b
Name: transmembrane protein 106B
Synonyms: 6430519M21Rik, 5830455K21Rik, 2310036D22Rik
Type: Gene
Species: Mouse
Chromosome: 6
Alteration at locus: Knockout
NCBI: 71900
Homologene: 56806
Genetic Alterations
Deletion of Tmem106b via flp and cre recombinase expression in mice carrying this allele to remove the lacZ sequence, neo selection cassette and the loxP-flanked critical exon(s).
ES Cell Line
Not applicable
Phenotype
Deletion of TMEM106B has limited phenotype on its own, but exacerbates accumulation of phosphorylated tau, accelerates neurological phenotypes, and worsens transcriptional signatures of disease in a mouse model of Alzheimer's/frontotemporal dementia.

Strain Development
The strain was derived from the germplasm containing the allele C57BL/6N-Tmem106btm2a(KOMP)Wtsi, which we used for IVF of C57BL/6J oocytes. Founder offspring were successively mated with beta-actin-Cre and beta-actin-Flp transgenic lines to delete the loxP-flanked exon 4 and the frt-flanked neo-lacZ cassette. We have bred out the Flp and Cre alleles and maintained the line on a C57BL/6J background by successive generations of heterozygous backcross to C57BL/6J wild-type mates. At least one generation included breeding Ht female with WT male to render the Y chromosome fully C57BL/6J.

MMRRC Genetic QC Summary
The MMRRC Centers have developed a genetic QC pipeline using MiniMUGA array genotyping to provide additional information on strain backgrounds for MMRRC congenic and inbred strains. For more information on when data may be available, or to request genotyping for a strain of interest, please contact csmmrrc@jax.org. Older strains may not have this information.

Colony and Husbandry Information

Cryo-recovered strains distributed by the MMRRC at JAX are shipped to the customer from the Pathogen & Opportunistic-Free Animal Room G200 - see https://www.jax.org/jax-mice-and-services/customer-support/customer-service/animal-health/health-status-reports.

Mice recovered from a cryo-archive will have health surveillance performed on recipient females. Health reports will be provided prior to shipment. If you require additional health status information, please email csmmrrc@jax.org.
