    Phenotype

    Homozygous and heterozygous mice have no ocular phenotype detected and no comprehensive morphological analysis.

    Conditional Phenotype: Crossing this floxed model with a global cre driver (bActin-Cre) to create a germline knockout of Armcx1 resulted in no ocular phenotype.

    Strain Development
    JM8A3.N1 mouse ES cells were electroporated using linearized Armcx1 targeting vector DNA. 336 neomycin (G418) resistant ES clones were picked and analyzed by PCR. ES clones #15, #30 and #212 were identified as correctly targeted at the Armcx1 locus by PCR and confirmed by Southern blot analysis. ES Clones #15 and #212 were selected for in vitro deletion of FRT floxed PGK-Neo cassette by electroporating pCAGGS-Flpe-Puromycin plasmid and selected for puromycin (1.5ug/ml) resistance. For ES clones #15 and #212, 44 and 37 clones respectively, were isolated and subjected to PCR analysis for absence of PGK-Neo cassette and tested for neomycin sensitivity. For ES clone #15 and #212, 13/44 clones and 11/37 clones respectively, were both negative for PGK-Neo cassette and neomycin sensitive. Two clones from each #15 and #212 PGK-Neo deleted were selected for karyotyping. ES cell lines were injected into blastocysts harvested after mating C57BL/6Hsd (Envigo) females to generate chimeric lines. Chimeras were backcrossed to test germline transmission.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
