The coding region of part of the cytoplasmic tail (FREFCIPTSSTIEQQNSARIRQNTREHPSTANTVDRTNHQ) within exon 3 was replaced with the sequence from the delta opioid peptide receptor (FRQLCRTPCGRQEPGSLRRPRQATTRERVTACTPSQ) and a floxed neo cassette. Expression of the receptor in homozygotes was similar to the endogenous receptor.

Strain Development
To generate the Oprm1tm1Jlw mice a ~12.5 kb SalI-SacI fragment of genomic DNA was used to generate the targeting vector. In this targeting vector, the wild-type MOP-R sequence was replaced with the rMOP-R sequence. A 2.1 kb cassette containing G418 (Fx-Neo) resistance flanked by loxP sites was inserted into a SpeI site in the intron downstream of exon 3. The targeting vector was linearized with SacI and transfected into embryonic E14 stem cells (derived from 129P2/OlaHsd) via electroporation. Clones were selected by G418 resistance. Homologous recombinants clones were confirmed by Southern blot analysis. A positive clone was injected into C57BL/6 blastocysts to create chimeric animals. F1 agouti progeny were genotyped and used to generate the Oprm1tm1Jlw knock-in mice.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
