Genetic Alterations
This strain harbors a duplication of the Gldc gene. Two loxP sites have been inserted flanking the Gldc gene, one using gene targeting in ES cells, the other with CRISPR/Cas-9 in zygotes. Mice carrying the two loxP sites in trans and a Hprt-Cre transgene were bred with wild type mice to identify mice that have undergone trans-allelic recombination, effectively duplicating the Gldc gene.

Phenotype
The Rln1-Gldc chromosomal segment is duplicated/triplicated in patients with schizoaffective disorder and bipolar disorder with psychotic features. This strain contains a duplication of the Gldc gene encoding the glycine decarboxylase gene, a glycine-degrading enzyme.
  • Homozygous mice: contain 4 copies of Gldc instead of the normal 2 copies
    • Glycine decarboxylase expression and activity in the brain are increased
    • miR-137 expression are increased (strongly associated with schizophrenia in GWAS) and Pyroxd2 expression
    • BDNF expression and Akt/mTOR signaling is decreased
    • Total glycine concentrations from hippocampal or prefrontal cortex is unaltered, the extracellular glycine concentration is reduced in dentate gyrus (DG) but not in CA1
    • LTP deficit at mPP-DG synapses, but not at CA3-CA1 synapses
    • Baseline and FCCP-stimulated mitochondrial respiration are reduced in DG but not in CA1
    • Behaviorally, mice display a startle habituation deficit, a latent inhibition deficit, working memory deficits in the Y maze and the water T maze, and both sociability and social preference deficits in the three-chamber social interaction test
  • Heterozygous mice: contain 3 copies of Gldc
    • Behaviorally, mice display a similar phenotype (with no deficit in the water T maze)

Strain Development
Two loxP sites flanking the Gldc gene were inserted into the mouse genome, one loxP site by gene targeting in v6.5 ES cells (which the authors report come from 129S1 mice), the other one by CRISPR/Cas-9 in C57BL/6 zygotes. Mice carrying both loxP sites and an Hprt-Cre transgene were generated and bred with C57BL/6J wild type mice. Offspring were screened for the presence of the Gldc duplication. In case the cre transgene was present, it was bred out. Selectable neomycin resistance markers that were included in the targeting vectors have been removed using Flp-FRT recombination. Mice were bred to C57BL/6J wild type mice for at least 8 generations. Mice were then inbred for 5 generations.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. 2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
