Genetic Alterations
This model harbors two mutations on different chromosomes.

Mir204tm1.1Dic : An IRES-lacZ reporter cassette as well as a loxP site were inserted 178 bp upstream miR-204 stem loop and a loxP site plus a neomycin selection cassette were placed 260 bp downstream miRNA stem loops by homologous recombination in ES cells. The chimeric mice were crossed to flippase-expressing transgenic mice to remove Frt or F3 (a mutant of Frt) flanked lacZ and Neo cassettes, leaving two loxP sites immediately flanking the miRNA stem-loops.

Mir211tm1.1Dic : An IRES-lacZ reporter cassette as well as a loxP site were inserted 171 bp upstream miR-211 stem loop and a loxP site plus a neomycin selection cassette were placed 365 bp downstream miRNA stem loops by homologous recombination in ES cells. The chimeric mice were crossed to flippase-expressing transgenic mice to remove Frt or F3 (a mutant of Frt) flanked lacZ and Neo cassettes, leaving two loxP sites immediately flanking the miRNA stem-loops.

Phenotype

Homozygotes are fertile, although the fertility is lower in female mice.

Conditional Phenotype: If the mice are crossed with flippase-expressing transgenic mice to remove the lacZ and Neo cassettes, and then crossed with CMV-Cre, the double knockout mice of miR-204 and miR-211 would exhibit accelerated aging, including hyper-mature cataracts with rupture, uveal melanosis, retinal dysplasia, follicular dysplasia, osteoarthritis, and accelerated Alzheimer's disease progression (in the background of APP/PS1).

Strain Development
The targeting vectors for miR-204 and miR-211 were constructed using the recombineering-based method.
An IRES-lacZ reporter cassette as well as a loxP site were inserted 178 bp upstream the miR-204 stem loop and 171 bp upstream the miR-211 stem loop. In miR-204, a loxP site plus a Neomycin selection cassette were placed 260 bp downstream the miRNA stem loops. For miR-211, they were placed 365 bp downstream the miRNA stem loops. Both lacZ and Neo cassettes are flanked by Frt or F3 (a mutant of Frt) sites. The constructs were linearized and introduced into W4 ES cells (originating from 129S6/SvEvTac) via electroporation. The targeted ES cells were selected with G418, confirmed with long-range PCR, and injected into C57BL/6J blastocysts to generate mouse chimeras. The strains of floxed miR-204 and miR-211 were backcrossed to C57BL/6 for over ten generations.

To generate the miR204/211 strain, the strains of floxed miR-204 and miR-211 were crossed with each other and the genotype of the offspring were confirmed with the PCR.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.
