Genetic Alterations
Enhanced Green Fluorescent Protein (EGFP) was inserted into the C terminal tail of the GPCR and IRES-Cre into the 3' UTR.
The EGFP modification allows for imaging for the gene at the protein level. The Cre modification allows expression mapping when crossed with Ai9 mouse. The Cre modification also allows a variety of other experimental techniques using Cre-dependent viral delivery of molecular tools.

    Strain Development
    Cas9 RNP was injected into zygotes with a homology donor to create the insertion. The mouse strain used to obtain the embryos was C57BL/6J (JAX #000664). The mouse strain used to implant the blastocysts was CD-1 (Charles River Laboratory). C57BL/6J zygotes were microinjected with 400 nM Cas9 protein, 20 ng/ul guide RNA and 50 ng/ul donor. In vitro transcribed, purified guide RNA was diluted in microinjection buffer (5 mM Tris–HCl pH 7.5, 0.1 mM EDTA), heated at 95°C for 3 min, and placed on ice prior to addition of Cas9 protein. The mixture was then incubated at 37°C for 5 min and placed on ice, after which the donor oligonucleotide was added. The final mixture was filtered through a 0.2 uM filter and held on ice prior to microinjection. Microinjected embryos were implanted in recipient pseudopregnant females. Resulting pups were screened by PCR and sequencing for the presence of the mutant allele. Independent male founders were mated to wild-type C57BL/6J females, and transmitted the mutant allele through the germline to offspring.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
