    Phenotype

    Srl flox mice are viable and are born in expected ratios.


    Conditional phenotype: When Srl is ablated in cardiomyocytes using a Tnnt2-Cre driver, it leads to a sex-biased cardiac phenotype. Male mice exhibit prolonged cardiac repolarization as a result, while female mice show normal characteristics compared to the control group.

    Strain Development
    Cas9 guide RNAs were designed to insert loxP sites flanking the 5’ end of Srl, encompassing the promoter and 1st exon. Selected guide RNAs were cloned into a T7-tRNA-guide RNA construct (UNC Animal Models Core) for T7 in vitro transcription of tRNA-gRNA fusion molecules. The in vitro transcribed tRNA-gRNAs were purified and eluted in microinjection buffer. The guide RNAs selected for embryo microinjection were Srl-5g63T and Srl-i1g40T. The donor plasmid included an 861 bp 5’ homology arm, 5’ loxP site inserted 385 bp 5’ of Srl exon 1, 620 bp critical region encompassing Srl promoter, exon 1, and 156 bp of intron 1, 3’loxP and FRT sites, and 1048 bp 3’ homology arm. Recombinant eSpCas9 protein was expressed in E. coli and purified. C57BL/6J zygotes were microinjected with eSpCas9 protein, 600 nM each tRNA-gRNA and 20 ng/ul donor vector in microinjection buffer. Injected embryos were implanted in recipient pseudopregnant females. Resulting pups were screened via PCR for the presence of the correct loxP insertions. Multiple founders with the Srl floxed allele were identified. Two founders were selected for sequence confirmation of the 5’ and 3’ loxP sites and mating to wild-type C57BL/6J animals for germline transmission of the floxed allele.

    1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
