Genetic Alterations

Two loxP sites and an frt-flanked neomycin resistance cassette were inserted flanking ~400 bp of the first coding exon 1 from 44 bp upstream through 358 bp downstream of the translation initiation codon. A tau-LacZ gene was also inserted 3' to the neomycin resistance gene.Exposure to Cre recombinase removes the floxed sequence creating a null allele.

Phenotype

Mice are healthy, fertile, and homozygous floxed mice are regularly obtained after intercrosses. Their numbers at weaning are reduced relative to Mendelian expectation, suggesting the unrecombined allele is hypomorphic. This hypothesis is supported by characterization of the floxed allele, which revealed that its expression was variably reduced relative to wild-type Smo.

Conditional phenotype: Cre-mediated recombination deletes the allele, recapitulating the Smo null phenotype. Mice develop shorter long bones when compared to wild-type littermates. Furthermore, chondrocyte differentiation proceeds normally but, proliferation of chondrocytes is reduced by about 50%.

Strain Development
A loxP site flanked targeting vector containing a 400bp fragment of exon 1 and an FRT-flanked neomycin resistance gene was utilized in the construction of this mutant. This construct, with a downstream tau-lacZ, was electroporated into 129X1/SvJ derived AV3 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts. The resulting male chimeric animals were crossed to Swiss Webster females. The strain was then maintained as a homozygote prior to sending to the Jackson Laboratory in 2002 (Strain #: 004526). The strain was then backcrossed to C57BL/6J for more than 10 generations by the Beier lab resulting in a line that is >99% C57BL/6J.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
