Phenotype
CLPTM1 (cleft lip and palate associated transmembrane protein 1) is a transmembrane protein involved with regulating GABAergic transmission and modulates inhibitory synaptic strength. Clptm1tm1a(KOMP)Wtsi knock-out (KO)-first mice contain a lacZ/neomycin cassette and a loxP-flanked exon 6. These mice can be converted to a Clptm1 conditional knock-out line by crossing to a FLP-recombinase.

Heterozygous mice are viable and fertile; homozygous mice are not viable. Clptm1tm1a(KOMP)Wtsi KO-first heterozygous mice have elevated surface synaptic GABAA receptors as well as elevated phasic and tonic inhibitory neurotransmission. This is associated with impaired long-term potentiation and contextual fear memory. These mice may be useful when studying GABAA receptor trafficking and inhibitory synaptic transmission.


Conditional phenotype: The line can be converted with Flp to a Clptm1 conditional knockout line.

Strain Development
C57BL/6N-derived JM8A3.N1 embryonic stem (ES) cells bearing the Clptm1tm1a(KOMP)Wtsi targeted allele were obtained from Knock-out Mouse Project (KOMP). The targeting cassette is composed of an FRT site followed by lacZ sequence, an initial loxP site, a neomycin resistance sequence, a second FRT site, and a second loxP site upstream of exon 6 of the cleft lip and palate associated transmembrane protein 1 (Clptm1) gene. A third loxP site is inserted downstream of exon 6. Targeted ES cells were injected into recipient blastocysts. The resulting chimeric mice were bred to C57BL/6J to confirm germline transmission. The Clptm1tm1a(KOMP)Wtsi knock-out (KO)-first mouse line was then backcrossed to C57BL/6J for 11 generations by the donating laboratory. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
