Genetic Alterations
Prop1neo has a lacZ-neomycin cassette replacing the start codon of exon 1 and parts of exon 2 of the Prop1 gene. These mice may be useful when studying pituitary development and hormone production.

Phenotype
Heterozygous mice are viable and fertile with no overt phenotype.

Homozygous mice are viable and fertile, but do demonstrate dwarfism, noticeable by 2 weeks of age.*

*The donating laboratory recommends delaying weaning of homozygous mice until ~5 weeks of age. These Prop1neo homozygous mice demonstrate phenotypes similar to the Prop1df allele, including stunted growth and absence of GH, prolactin and TSH. Genetic background affects the degree of respiratory distress and viability of homozygous mice with ~6.7% surviving to 2 weeks of age on the 129S1 background. After backcrossing the 129S1 homozygous mice to C57BL/6 for 4 generations, viability was increased to ~17.5% homozygous surviving to 2 weeks of age. The donating laboratory maintains these mice on a mixed B6;129S background. It is recommended that this line be maintained on a mixed B6;129 genetic background.

Strain Development
The Prop1neo targeting vector containing a lacZ fused to a PGK-neomycin (neo) cassette was inserted in-frame with the initiation codon in exon 1 of the paired like homeodomain factor 1 (prop1) gene, deleting all of exon 1, intron 1, and part of exon 2. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl+-derived R1 embryonic stem (ES) cells and correctly targeted ES cells were injected into C57BL/6 blastocysts. Chimeric mice were bred to C57BL/6J mice to confirm germline transmission. The Prop1neo mouse line was maintained on a mixed B6;129 background by the donating laboratory.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
