    Strain Development
    To create the targeting vector PSN7, two fragments of the Tcof1 gene were PCR-amplified, cloned, and sequenced. A loxP site was inserted upstream of exon 1, and the modified fragments were combined in the pTKLNCDL vector to generate PSN7. Sanger sequencing confirmed the desired direction and correct junction sequences. The PSN7 construct was linearized and 20 µg of DNA was electroporated into 129/SvEv ES (HZ2.2) cells. Three correctly targeted cell lines were identified by PCR and Southern analysis. To generate the conditional allele, one of the correctly targeted cell lines (1A5) was transiently transfected with Cre recombinase. Two clones that contained a deletion of the neo/CD cassette were used to generate the conditional knockout mice. Non-surviving clones were expanded and genotyped for the conditional allele by PCR and Southern analysis. Cell lines were karyotyped prior to blastocyst injections. To analyze the mouse model on a B6/129 background, Wnt1-Cre transgenic mice (Swiss Webster) were crossed with C57BL/6 mice for at least 10 generations. Then hemizygous Wnt1-Cre mice were mated with homozygous conditional allele mice to generate conditional heterozygous knockout mice. Conditional heterozygous knockout males were then used to mate with homozygous floxed Tcof1 females to generate homozygous conditional knockout mice.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary.     2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
