Genetic Alterations
Mice harboring a floxed allele of Zc3h14 were bred to a cre-expressing strain to remove the inserted cassette and exon 13 of the Zc3h14 gene, resulting in a knockout allele.

Phenotype
Homozygous mice are phenotypically normal. Other groups have reported that Zc3h14 (Msut2) knockout mice have cognitive defects and enlarged brain ventricles, these phenotypes are not observed in these mice, indicating that this strain is unique.

Conditional phenotype: When in combination with a tau transgene that causes neurodegeneration, transgenic PS19 mice overexpressing tau and lacking the Zc3h14 (Msut2) gene exhibited decreased learning and memory deficits, reduced neurodegeneration, and reduced accumulation of pathological tau compared to PS19 tau transgenic mice expressing Zc3h14 (Msut2).

Strain Development
B6-Zc3h14tm1a(KOMP)Wtsi ES cells were used for microinjection and the chimera were mated to C57BL/6J for germline derived F1 mice. F1 mice were mated to C57BL/6J for colony expansion. Heterozygous offspring were mated to produce homozygotes, and the strain was subsequently maintained as a homozygous line. B6-Zc3h14tm1a animals were crossed with a Flp expressing strain (129S4/SvJaeSor-Gt(ROSA)26Sortm1(FLP1)Dym/J), resulting in a floxed conditional allele. Mice with the conditional allele were then crossed with a cre-expressing strain (B6.129S4-Meox2tm1(cre)Sor/J) to remove exon 13 of the Zc3h14 gene as well as all inserted transgene sequences. This strain was then backcrossed to C57BL/6J for at least eight generations.
