Phenotype

Loss of H19 results in progressive cardiomyopathy. In adulthood, H19-deficient mice (~1-year old mice) display the following phenotypes:

  • Cardiac hypertrophy and fibrosis
  • Elevated cardiac failure markers (ANP, Myh7, cleaved Caspase-3, cleaved PARP)
  • Abnormal echocardiography phenotypes (dilated ventricles, reduced ejection fraction, thickened walls, abnormal flow)
  • Low blood pressure and abnormal arterial structure/function
  • Endothelial dysfunction with increased frequency of endothelial to mesenchymal transition (EndMT)

Strain Development
A cre-loxP-based deletion strategy was for generating H19DeltaExon1 mice. The targeting vector carried an 11.7-kb BglII fragment with H19 sequences from between -2 and +9.7 kb (all base pairs are described relative to the H19 transcriptional start site). A loxP-flanked cassette with neomycin resistance and thymidine kinase genes (neo-tk) was inserted at DraIII (3 bp), and an additional loxP site was inserted at BsmI (710 bp).
The diphtheria toxin-A gene was inserted for negative selection. The linearized vector was electroporated into mouse RI embryonic stem cells derived from the (129X1/SvJ x 129S1/Sv)F1-Kitl+ strain. Correct clones were identified by Southern hybridization and then re-electroporated with pBS185 (Invitrogen) to direct excision of neo-tk, and the excision was detected by Southern hybridization. Correct clones were injected into C57BL/6J blastocysts to generate chimeric founder mice that were mated with EIIa-cre transgenic females (JAX strain 003724) to generate strains deleted for exon I. The exon I excision was detected by PCR and confirmed by sequencing. H19DeltaExon1 were then generated by backcrossing heterozygouse females with C57BL/6J (JAX strain 000664) males for at least 10 generations.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.
