Genetic Alterations
In this allele, the H19 gene carries two targeted deletions (approximately 40 bp each) that remove the binding sites for let-7 microRNAs.
The resulting mutant RNA is expressed at wild-type levels and preserves several normal functions, including regulation of Igf2.

Phenotype

H19ΔLet7 mice are viable and fertile. Mice carrying the H19ΔLet7 mutation express H19 at wild-type levels. Starting at one year of age, H19ΔLet7mice displayed cardiomegaly as measured by a 32% increase in heart weight/tibia length ratios. Transverse sections suggested hypertrophy, which was quantified as increased fiber diameter. Cardiac hypertrophy in H19ΔLet7 mice was accompanied by increased interstitial and perivascular fibrosis. The pathologic nature of the observed cardiac myopathies in these mutant mice was confirmed by the increased levels of cardiomyopathy markers.

H19ΔLet7lacks the growth defects associated with complete ablation of H19 since regulation of Igf2 by the mutant RNA is normal.

Strain Development
The H19ΔLet7 allele was generated using CRISPR/Cas9 gene editing of R1 mouse embryonic stem cells (ESCs). Guide RNAs were used to delete 25 bp in exon 1 and 48 bp in exon 4. Positive clones were identified by PCR screens and then confirmed by sequencing 686 bp spanning the exon 1 deletion and 1086 bp spanning the exon four deletion. Founder mice were obtained by injecting mutated ESCs into C57BL/6J blastocyts and then backcrossed twice with C57BL/6J females
The strain has been backcrossed to C57BL/6J for at least 10 generations.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
