Genetic Alterations
A knockout-first allele (tm1a) of Usp20 was sequentially subjected to Flp and Cre recombination, generating a knockout mouse in which exons 7 and 8 were deleted. Residual FRT and loxP are present between exons 6 and 9 of Usp20.

    Usp20 knockout mice are born at expected rates and are phenotypically normal under normal conditions.

    Conditional Phenotype:
    • Under pressure overload, mice develop left ventricular hypertrophy.
    • Upon IL-1β stimulation, endothelial cells (EC) from Usp20 knockout mice exhibited increased phospho-p65 protein levels compared to wild-type littermates.
    • Ex-vivo angiogenesis assays such show significantly increased EC sprouting in both male and female Usp20 knockout mice compared to their WT littermates (p < 0.05).

    Strain Development
    The L1L2_Bact_P cassette was inserted at position 30893994 of Chromosome 2 upstream of the critical exon(s) (Build GRCm39). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by a neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of the targeted exon(s) at position 30895294. The critical exon was thus flanked by loxP sites.

    Mice (with a tm1a allele) were mated with B6.129S4-Gt(ROSA)26Sortm1(FLP1)Dym/RainJ (RRID:IMSR_JAX:009086) to remove the lacZ and neo cassettes of the “knockout-first” allele. Thereby restoring Usp20 expression and rendering the Usp20 allele “floxed” with loxP sites flanking critical exons. Floxed Usp20 mice were then bred with CMV-Cre mice (background not specified) to generate knockout mice (Usp20-/-). Knockout mice were backcrossed to C57BL/6J, and heterozygous mice (Usp20-/+) were bred to obtain homozygous Usp20+/+ wild-type and Usp20-/- littermates.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

    3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
