Genetic Alterations
A conditional by inversion (COIN) intron (as described in PMID: 23918385), consisting lox66_SA-Egpf-polyA_lox71, was inserted in the antisense orientation into intron 3 via homologous recombination, splitting intron 3 into 2 new exons (94 and 2,258 bp in length). The two lox sites are in reverse complementation orientation, thus cre-mediated recombination will irreversibly invert the inserted sequence. In addition, an FRT flanked drug selection cassette (hygromycin- or neomycin-delta-thymidine kinase cassette) was inserted after the COIN intron.

Mice were bred to Flp deleter mice (B6.129S4.Gt(ROSA)26Sortm1(FLP1)/Dym/RainJ) to excise the Neomycin delta thymidine kinase cassette.

Breeding with Cre-expressing line results in flipping of the splice acceptor, deletion of the normal Sox10 transcriptional activation domain and expression of a JRed reporter

Conditional Phenotype: When crossed with a neural crest specific Cre driver, heterozygous offspring exhibit hypopigmentation and gut motility defects. It is recommended that the line should be maintained as heterozygotes.

Strain Development
The Sox10-COIN construct was generated by incorporating a Neo drug selection cassette driven by the human UB(c) promoter flanked by FRT sites as well as an inversion cassette containing a splice acceptor site linked to the red fluorescent protein JRed followed by a rabbit B-globin polyadenylation sequence. This 'COIN' cassette was introduced into a Sox10 BAC clone RP23 28O11 by in vitro homologous recombination in E. coli that resulted in targeted integration downstream of Sox10 exon 4. The targeting vector was linearized and electroporated into JM8.F6 ES cells derived from C57BL/6N (Pettitt et al., 2009) which were subsequently subjected to G418 selection. 480 clones were picked into individual wells of 96-well plates and were then screened by PCR for integration of the JRed reporter. PCR-positive clones were subsequently confirmed by Southern blotting. ES cell clones were introduced into C57BL/6J-TyrC-2J/J blastocysts. Chimeras were bred for transmission of black offspring and subsequent progeny were bred with Flp deleter mice (B6.129S4.Gt(ROSA)26Sortm1(FLP1)/Dym/RainJ) to excise the Neomycin delta thymidine kinase cassette. Progeny lacking the Neo cassette were bred with C57BL/6J mice to make Sox10COIN lines congenic on this inbred strain background.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
