Genetic Alterations
CRISPR-targeting introduced a 59-bp deletion (D78Vfs*6, D70Vfs*6, D70Vfs, ZF1a+59) in exon 3 of the Zfp423 gene, creating an early frameshift, removing one side of a predicted stem-loop in the mRNA.

Phenotype

Zfp423em12Haml mice (D78Vfs*6) exhibit a hypomorphic, allele-specific phenotype that is milder than other premature termination codon (PTC) alleles of Zfp423.

  • Homozygous mice:
    • Show reduced body weight, cerebellar vermis hypoplasia, and mild to moderate ataxia
    • Display partial postnatal viability and less severe neuroanatomical defects
    • This allele produces a detectable truncated Zfp423 protein, resulting in residual protein expression at approximately ~50% of wild-type levels
  • Heterozygous mice:
    • Display mild haploinsufficiency, including modest reductions in vermis size and body weight

Conditional phenotype: The phenotype is constitutive and is more severe on C57-related genetic backgrounds.

Strain Development
The Zfp423em12Haml (D78Vfs*6) strain was generated using CRISPR/Cas9 genome editing. Guide RNAs were designed using public web-based tools to minimize off-target effects. Genome editing was performed by microinjection of one-cell stage embryos. The D78Vfs*6 mutation was originally generated in C57BL/6N zygotes and subsequently crossed to FVB/NJ for at least ten generations using flanking markers to minimize the congenic interval to D8Mit45-D8Mit51.

1 The distribution fee covers the expense of rederiving mice from a live mouse; you will receive the resulting litter. The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
