Phenotype
C1qtnf5−/− mice lack both C1qtnf5 transcript and protein. Knockout mice exhibit a significantly increased number of fundus autofluorescent spots compared with wild-type mice by ~19 months of age. Early retinal pigment epithelium (RPE) stress is evident as early as 6 months, characterized by RPE thinning and vacuolization, and is followed by late-onset retinal degeneration. Ultrastructural analyses reveal progressive accumulation of basal laminar sub-RPE deposits beginning around 11 months of age and increasing with age.

Strain Development
To generate C1qtnf5+/+ mice, 11.5 kb of mouse genomic DNA fragment containing exons 1–3 of Ctrp5 (C1qtnf5) was cloned from a positively identified C57BL/6 (RPCI-23) BAC clone. The LoxP/FRT flanked Neo cassette was inserted 693 bp 3′ to the C–>G point mutation engineered into exon 3 using a Red/ET recombination kit. Overlap extension PCR was used to generate the point mutation. The single loxP site was 5′ to exon 2. Targeting vector was linearized and electroporated into C57BL/6 embryonic stem cells which were subjected to selection with G418 antibiotic and ganciclovir. Correctly targeted embryonic stem cells were injected into BALB/c blastocysts and implanted into pseudopregnant mice, to generate chimeras. These chimeras were mated with C57BL/6J mice to transmit the targeting allele.

To generate homozygous C1qtnf5 knock-out (C1qtnf5−/−) mice, C1qtnf5+/+ mice were crossed with global Cre mice on the same C57BL/6J background to remove exons 2 and 3 of the C1qtnf5 gene.
