A conditional allele of Tgfbi was generated by flanking exon 3 with loxp sites. Cre-mediated recombination excises exon 3, resulting in disruption of Tgfbi gene function.

Phenotype

Tgfbi conditional mice are viable and do not exhibit an overt phenotype under baseline conditions.

Conditional Phenotype: Upon Cre-mediated recombination, excision of exon 3 results in loss of Tgfbi expression in macrophages (MΦs). MΦ-specific deletion of Tgfbi inhibits polarization toward the M2 phenotype and reduces MΦ-mediated stimulation of pancreatic cancer growth. This deletion significantly enhances anti-tumor immunity, increases tumor-infiltrating CD8+ T cells, reduces T cell exhaustion, and sensitizes pancreatic tumours to chemotherapy. These effects are associated with regulation of extracellular matrix and immune-related factors, including fibronectin 1, Cxcl10, and Ccl5.

Strain Development
For generating Tgfbiflox/flox mice via CRISPR-Cas9 gene editing, three week-old female C57BL/6 mice as embryo donors and ten-week-old stud male mice as sperm donors were obtained from Jackson Laboratory. Using the online CRISPR RGEN Tools, sgRNAs flanking exon 3 of Tgfbi were designed and synthesized. A repair template was designed to introduce the desired loxP sites flanking exon 3 of Tgfbi. A mix containing the sgRNAs, Cas9 protein, and lssDNA was microinjected into zygotes, which were subsequently surgically transferred to pseudopregnant CD-1 surrogate females.
The mutant allele was confirmed by PCR and DNA sequencing across both homology arms and both engineered loxP sites. Tgfbi PCR primers were used to amplify across the entire targeted region, after the correctly targeted allele was confirmed by DNA sequence, animals were further genotyped for the presence of the engineered allele via PCR. Founder mice were mated to C57BL/6J breeders, and derived N1 progeny were identified by PCR and sequencing. N1 were then mated again to C57BL/6J breeders to generate N2 mice identified by PCR and sequencing. N2 heterozygous mutant mice were mated for production of homozygous cohorts.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. 2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. 3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
