Genetic Alterations
The transgenic construct contained a 4.8 kb fragment of Pdgfrb including the promoter and the first exon fused to a nuclear-targeted cre recombinase sequence. The construct was injected into fertilized oocytes. Founders were crossed to Gt(ROSA)26Sortm1Sor reporter mice to analyze Cre activity in embryonic and adult tissues. Line 35 was used for further experiments.

Phenotype

Tg(Pdgfrb-Cre)35Vli transgenic mice display no overt phenotype under baseline conditions.

Conditional phenotype: When crossed with the Gt(ROSA)26Sortm1Sor strain,Cre-mediated recombination (b-galactosidase activity) closely overlaps with endogenous Pdgfrb expression in vascular mural cells, smooth muscle, mesangial cells, dermal fibroblasts, myocardium, and embryonic mesenchyme.

When used to drive expression of a conditional myc-tagged Cthrc1 transgene, Cre-driven expression similarly recapitulates endogenous Pdgfrb distribution. However, following carotid artery ligation, dedifferentiation of vascular mural cells show reduced ROSA26 reporter activity despite continued Cre-dependent transgene expression, indicating suppression of ROSA26 reporter activity in remodeling vessels rather than a loss of Cre function.

Cre-positive parents should only be bred with wild-type mice.

Strain Development
To generate the construct, a -4.7/+0.1 kb fragment of the Pdgfrb promoter including the first exon was amplified from mouse genomic DNA. The amplified sequence was fused to a nuclear targeted Cre recombinase sequence. Oocytes from an FVB/N female mated with a homozygous male of the B6;129S4-GT(ROSA)26Sortm1Sor strain (JAX #003309) were injected with the construct. Founder animals were identified and crossed to Gt(ROSA)26Sortm1Sor mice for analysis. Founder #35 was selected for further breeding, expansion, and was mated to FVB females. The resulting progeny were screened for presence of the transgene. The strain was then backcrossed for at least 10 generations to C57BL/6J (JAX #000664). Mice were then sibling mated after the backcrossing was complete, for at least 5 generations (F5+).

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
