    Phenotype

    R26-A1bg-3HA knock-in mice exhibit no detectable phenotype in the absence of Cre-mediated recombination.

    Conditional phenotype: Following cardiomyocyte-specific activation via crossing with STOCK Tg(Tnnt2-cre)5Blh/JiaoJ (RRID:IMSR_JAX:024240), offspring are are viable, fertile, and recovered at Mendelian ratios. No overt structural or functional cardiac abnormalities were observed. Expression of the A1BG-3xHA fusion protein was confirmed by immunoblotting with anti-HA antibodies, and the tagged protein localizes to the cardiomyocyte extracellular surface. The knock-in allele enables immunopurification of native A1BG-containing complexes without disrupting cardiac morphology, ventricular structure, or electrophysiological function.

    Strain Development
    A Cas9 guide RNA targeting the first intron of the Gt(ROSA)26Sor (Rosa26) locus was cloned into a T7 promoter vector, transcribed in vitro using T7 RNA polymerase, and purified. A donor plasmid was constructed containing Rosa26 homology arms flanking a neomycin resistance cassette, CAG promoter, LoxP-STOP-LoxP cassette, mouse A1bg cDNA fused to a C-terminal 3xHA tag, Woodchuck Hepatitis Virus Posttranscriptional Regulatory Element (WPRE), and a rabbit beta-globin polyadenylation sequence.

    Recombinant Cas9 protein was expressed in E. coli and purified.The C57BL/6N-PRX-B6N #1 ES cell line was nucleofected with 1 μM Cas9 protein, 1.2 μM guide RNA, and 20 ng/μl supercoiled donor vector. G418-resistant clones were screened by PCR and correctly targeted clones were confirmed by Southern Blot analysis. Two validated clones were microinjected into Albino C57BL/6 blastocysts to generate chimeric mice. Chimeras were bred to Albino C57BL/6N females for germline transmission of the targeted allele. ES cell clone E2 achieved germline transmission of the targeted allele. Heterozygous F1 animals (R26R-A1BG-3HA+/−) were bred to wild-type mice, and the genotypes were confirmed by sequencing and PCR.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.
