Genetic Alterations
This strain carries a CRISPR/Cas9 generated compound heterozygous mutation in the Tulp3 locus with the following two alleles:
  • A splice-site hypomorphic allele (i4), which alters the intron 4 splice acceptor and reduces Tulp3 expression
  • A null deletion allele (Δ5), which deletes exon 5 and eliminates TULP3 function

Phenotype
Tulp3-i4/Δ5 mice exhibit genotype-dependent reductions in TULP3 expression, with heterozygous animals expressing ≤5% of wild-type levels. Both heterozygous and homozygous mice show increased weight gain over time relative to their littermate controls, as well as modest brain overgrowth. This includes an approximately 5–6% increase in sagittal vermis area and anterior–posterior dimension of the cerebellar hemispheres, with heterozygotes also showing nominal evidence of increased brain width (2.6%).

In addition, mice develop progressive cystic kidney disease, characterized by an age-dependent increase in cystic index and associated ciliopathy features, including shorter and fewer primary cilia.

Strain Development

This strain was generated by CRISPR/Cas9-mediated genome editing in FVB/NJ one-cell embryos using guide RNAs, cas9 protein, and donor pooled Ultramer oligonucleotides to modify splice regulatory regions and coding sequences of Tulp3. Edited embryos were implanted into pseudo-pregnant females. Diluted editing mixes as injected contained crRNA:tracrRNA, Cas9, and pooled Ultramer oligonucleotides. Tail biopsy DNAs from potential founder (G0) animals were used for PCR-based sequencing. Edited lines were maintained as coisogenic by serial backcross to FVB/NJ. Genotypes for stock maintenance and experimental subjects were determined by PCR assays.
