Phenotype
Homozygous and heterozygous mice do not exhibit overt abnormalitites and are phenotypically wild-type under baseline conditions.

Conditional Phenotype: Upon Cre-mediated recombination, exon 2 is deleted resulting in a loss of Alx1 function. Conditional inactivation of Alx1 in specific lineages leads to developmental defects, including absence of extraocular muscles and craniofacial abnormalities.

Cranial mesoderm-specific Alx1 inactivation resulted in complete extraocular muscle (EOM) agenesis accompanied by failure of activation of the core myogenic regulatory network specifically in, and increased apoptosis of, the EOM progenitor cells.

Temporally induced Alx1 inactivation demonstrated that EOM myogenesis requires Alx1 function before formation of the EOM primordium. This identifies ALX1 as a unique and specific upstream regulator of EOM myogenesis, and provides insights into pathogenic mechanisms underlying ALX1-type frontonasal dysplasia, as well as molecular mechanisms controlling cell fate specification in the early cranial mesoderm.

Strain Development
Alx1 conditional mice were generated using CRISPR/Cas9-mediated genome editing in C57BL/6J zygotes. Guide RNAs targeting introns 1 and 2 were used to introduce loxP sites flanking exon 2. Initial founder mice only transmitted the correctly integrated 5' loxP site in the intron-1 region to the G1 generation. Heterozygous G1 male mice were crossed to C57BL/6J inbred females and the resultant G2 heterozygous carriers were intercrossed to generate mice homozygous for the 5' loxP insertion. Zygotes from the homozygous intercrosses were retargeted to insert the 3' loxP site in intron 2 and founder mice were crossed to C57BL/6J inbred mice to generate mice heterozygous for the Alx1 conditional allele carrying the correctly integrated loxP sites in both intron 1 and 2, verified by Sanger sequencing. Subsequent generations of Alx1 conditional mice were maintained on the C57BL/6J inbred background.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.

3 An aliquot is one straw or vial with sufficient sperm to recover at least one litter of mice, as per provided protocols, when performed at the MMRRC facility. The MMRRC makes no guarantee concerning the success of these procedures when performed outside the MMRRC facilities.
