Phenotype

BRD4 is a transcriptional and epigenetic regulator that plays a pivotal role during embryogenesis and cancer development.

  • Brd4 heterozygous mice
    • Produce viable, fewer offspring but are otherwise relatively normal, aside from metabolic defects
    • Adipose tissue shows reduced expression of Adipoq and fatty acid synthesis related genes
    • Decreased circulating ADIPOQ protein levels compared to wild-type mice at 21 days after birth
  • Brd4 homozygous mice
    • Embryotic lethal

In vitro, BRD4 depletion by shRNA or inhibition with the BET inhibitor (+)-JQ1 in 3T3-L1 adipocytes leads to reduced Adipoq expression and lipid droplet accumulation. This is accompanied by decreased expression of insulin sensitivity–related genes, as well as reduced P-TEFb recruitment and histone acetylation at the Adipoq gene.

Strain Development
BAC clones containing the murine Brd4 locus were isolated to construct a targeting vector. This vector contained an En-2 splice acceptor fused to a β-geo reporter and polyadenylation (pA) cassette, flanked by 7.5 kb (5′) and 3.4 kb (3′) homologous regions. The construct was electroporated into murine 129 R1 embryonic stem (ES) cells (derived from (129X1/SvJ x 129S1/Sv)F1-Kitl+)), and recombinant clones were selected using G418. Clones with successful disruption of the Brd4 gene were identified by Southern blot analysis using probes external to the targeting construct. Homologous recombination resulted in replacement of exon 3 with the selection cassette. Recombinant ES cells were injected into blastocysts to generate chimeric mice, and animals with germline transmission were subsequently backcrossed to C57BL/6 mice for at least 10 generations.
