Genetic Alterations
Insertion of a CC042/GeniUnc-derived genomic interval containing the Tip3 QTL on chromosome 15 in a C57BL/6J genetic background. Tip3 was mapped to chromosome 15 with a peak at 83.1 Mb and a Bayesian interval of 53.7–89.7 Mb.

    Phenotype

    The phenotype expected for this strain is based on the Tip3 QTL identified in the CC001 × CC042 genetic mapping study. In the study, Tip3 was significantly associated with lung IFN-γ levels following M. tuberculosis infection and explained an estimated 13.5% of the phenotypic variance in the mapping population. The CAST-derived haplotype at Tip3 was associated with lower IFN-γ production, while the alternate 129-derived haplotype was associated with higher IFN-γ levels.

    Strain Development
    The C57BL6J.CC042 congenic strain was developed to isolate the Tip3 QTL identified through genetic mapping of tuberculosis-associated immune phenotypes in a CC001/Unc × CC042/GeniUnc F2 population. The original mapping study identified Tip3 on chromosome 15, with a Bayesian interval of 53.7–89.7 Mb and a peak at 83.1 Mb.

    A genomic region from CC042/GeniUnc containing Tip3 was introduced onto the C57BL/6J background by congenic backcrossing to nine generations. Selection during strain development was guided by MiniMUGA array genotyping.

    MiniMUGA genotyping performed in early 2024 defined the retained CC042-derived chromosome 15 interval as approximately 3–93.3 Mb. Thirty-two MiniMUGA markers within this interval are diagnostic between C57BL/6J and CC042.

The litter will contain at minimum one mutant carrier; the actual number of animals and the gender and genotype ratios will vary. (Typically, multiple breeder pairs can be established from the recovered litter.) Prior to shipment, the MMRRC will provide information about the animals recovered. If you anticipate or find that you need to request specific genotypes, genders or quantities of mice in excess of what is likely from a resuscitated litter, you may discuss available options and pricing with the supplying MMRRC facility.

    2 An aliquot contains a sufficient number of embryos (in one or more vials or straws and based on the transfer success rate of the MMRRC facility) to transfer into one to three recipients. The MMRRC makes no guarantee concerning embryo transfer success experienced in the recipient investigator's laboratory. Neither gender nor genotype ratios are guaranteed.
